ABSTRACT

Objective@#To assess the value of detecting multiple rearrangements of MLL gene in children with acute mononuclear leukemia (AML).@*Methods@#Eighty six children with AML were analyzed by fluorescence in situ hybridization (FISH), chromosomal karyotyping and multiplex reverse transcription-PCR (RT-PCR).@*Results@#Cross signals were detected by FISH in 26 cases, and 30.2% were detected with MLL gene rearrangements. R-band karyotyping analysis revealed 14 translocations with breakages involving 11q23 and 5 other aberrations, which yielded an overall detection rate of 22.1%. Multiple RT-PCR has detected 12 fusion genes produced by the MLL translocation, which yielded a detection rate of 14.0%. A significant difference was found in the detection rate of the three methods (P<0.05).@*Conclusion@#Combined use of FISH, chromosomal karyotyping and multiplex RT-PCR can improve the detection of MLL gene rearrangements and provide important clues for clinical diagnosis, treatment and prognosis of AML.

ABSTRACT

Objective To find out the impact of SHI-1 cells on VOCs in the headspace, and to assess the feasibili-ty of VOCs analysis in leukemia patients or with other tumors in the differential diagnosis by determing VOCs in the headspace of acute mononuclear leukemia cells(SHI-1). Methods The air samples from the headspace of SHI-1, Human macrophages cells and medium control were collected by syringes, and then determined by means of solid phase microextraction-gas chromatography/mass spectrometry ( SPME-GC/MS ) in order to have a better under-standing of the concentration distributions and changes of VOCs in SHI-1 cells' headspace. Results Using Mann-Whitney U test, we found that eight kinds of VOCs were different among them. They were 2,4-dimethyl heptanes, 4-methyl octane, chloroform, benzene, 3,7-dimethyl dodecanese, hexanol, cyclohexanol, cetane. And alkane, benzene were significantly higher than the control group ( P<0.05 ) , alcohol decreased significantly ( P<0.05 ) . Conclusion SHI-1 cells have an impact on the concentration distributions of VOCs in their headspace. The most outstanding VOCs are alkanes, alcohol and benzene compared with the blank control group. They may be potential markers characteristic of leukemia cells.

ABSTRACT

OBJECTIVE:To evaluate the economic effectiveness in different chemotherapeutic schemes for acute mononu?clear leukemia(M 5 ).METHODS:Using pharmacoeconomic cost-effectiveness analysis,three chemotherapy schemes for M 5 we_ re compared.RESULTS:Among three schemes,DAE scheme was the best one.CONCLUSION:The results show that phar?macoeconomics plays an important role in optimizing therapeutic scheme,in helping the physician to choose the best therapeutic plan in making clinical decision.
